Preamble
This lab-based course introduces modern tools of molecular cloning; protein expression and
purification and is designed for upper level undergraduate students majoring in Biology.
Students learn how to apply a range of recombinant DNA tools to modern biochemical
research.

Syllabus
Cloning and Protein Expression Laboratory
Cloning of DNA fragments: Cloning of gene by PCR method; Restriction Digestion of the
gene; Ligation and Transformation of the gene into a plasmid; Plasmid DNA preparation;
Verification of isolated plasmid DNA;
Expression of protein: Preparation of competent cells; Transformation of clone into
competent cells; Growth and induction with IPTG; Harvesting and preparation of cell lysate;
SDS-Gel electrophoresis
