Preamble
Tissue Culture, Immunofluorescence, Basic Fluorescence Microscopy and Image Analysis

Syllabus


  • Tissue Culture: Media Preparation, Trypsinization and Culturing eukaryotic cells; Hemocytometer counting of cells; Cryo-preservation of cells;

  • Immunofluorescence: Using Primary and fluorescently labelled secondary antibody to localize protein distribution in cells;

  • Basic Fluorescence Microscopy: Measuring Point Spread Function, Resolution, Scale Calibration, camera-based Image acquisition;

  • Image Analysis: Basic Image Analysis with FiJi/Matlab Understanding Digital Images; Object Detection; Image registration; Introduction to coding for Image analysis

  • Short project: Design an imaging-based short experiment and use image analysis tools to extract information.

  • Basic Flow cytometry: Basic introduction to Flow Cytometer. Understanding the principles and working of a Flow Cytometer. Designing a simple experiment that will demonstrate how two lineages of cells can be counted and separated in a Flow Cytometer.
